Bioavailability Enhancement of Artemether and Lumefantrine by Improving Solubility and Dissolution Rate using Solid Dispersion Technique

Artemether-lumefantrine is World Health Organization approved fixed dose combination for malaria treatment. These drugs have poor bioavailability due to low solubility and dissolution. The objective of this work was improve by increasing dissolution both drugs. In solid dispersion using hydrophilic carriers like polyvinylpyrrolidone K-30, Soluplus Lutrol F68 solvent evaporation technique prepared evaluated solubility, flow property, differential scanning calorimetry, x-ray diffraction analysis, electron microscopy, disintegration study. were determined simultaneously high performance liquid chromatography method various surfactants in different media. Solubility the increased all dispersions compared pure Flow properties containing K-30 better than other time immediate release tablet drug (1:0.2) less dispersions. X-ray calorimetry microscopy results indicated decrease crystalline nature their rate enhanced plain marketed formulations acidic buffer myrj 52 (1 %) as medium. pharmacokinetic studies mice revealed that artemether-lumefantrine had higher area under curve, maximum plasma concentration artemether lumefantrine tablet.
